Effects of gluten-free diet on immune parameters in Parkinson's disease

Tato studie zkoumá možné, i malé, vlivy bezlepkové diety v porovnání se standardní dietou na rozvoj Parkinsonovi nemoci s využitím 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridinem (MPTP) indukovaném myším modelu Parkinsonovi nemoci, a samců kmene C57Bl6. Tento výzkum zahrnuje zavedení jak akutního tak chronického myšího MPTP modelu, a řadu navazujících flow cytometrických analýz zaměřených na regulační T buňky, cytokiny - interleukin 10 (IL-10) a interferon gama (IFN-), gamma/delta T buňky, a NK buňky ve slizničních a sytémových lyfatických orgánech. Pro posouzení vlivu na chování jse použili test otevřeného pole, který nám umožnil posoudit lokomotorickou aktivitu a explorativní chování myší v závislosti na podávané dietě. Dále jsem použili také imunofluorescenční barveni pro validaci efektu MPTP, které nám poskytlo vizuální potvrzeni neuroanatomických změn indukovaných neurotoxinem. První výsledky naznačují malé ale slibné známky pozitivních efektů bezlepkové diety. Nicméně tyto změny lze pouze opatrně interpretovat, protože je potřeba opakovaných a podrobnějších experimentů k posouzení zazanmenaných efektů. Screening of probiotics bacterial strains aimed at proof of gluten degradation

No other treatment for celiac disease and other gluten-related disorders has been found so far except for a lifelong diet, which is significantly restrictive for the patient. Moreover, it is not always possible to adhere strictly to this diet, mainly due to random gluten ingestion, which leads to health complications associated with these diseases. Currently, vaccination is being tested as an alternative treatment, and there is also increasing attention paid to enzymes that could break down the immunogenic part of gluten, specifically in its gliadin component. This thesis examined bacteria that could potentially break down this sequence in gliadin and thereby reduce its concentration during detection. A considerable proportion of the tested bacteria belong to probiotics to ensure their future use for humans is safe. To obtain data, the fact was utilized that as bacteria grow, the turbidity increases proportionally, which was subsequently detected to determine the amount of bacterial growth. The RIDASCREEN® Gliadin competitive kit was used for the gliadin detection test, which works on the principle of a competitive ELISA method with R5 antibodies. The result of the study was the finding that only the bacteria Bacillus subtilis was able to break down gliadin among the examined bacteria. It was also measured how much gliadin the bacteria could break down at different concentrations, and according to the data, it was evaluated that it is appropriate to adapt the culture to the presence of gliadin for a greater amount of broken gliadin.
Awareness of general nurses about the treatment of celiak disease

Abstract Objective: The bachelor's thesis deals with the awareness of general nurses about the treatment of celiac disease. The first chapter of the theoretical part is focused on the digestive system, small and large intestine. The following chapter is devoted to the definition of celiac disease, its history and clarification of the concept of gluten. The next chapter presents the symptoms, diagnosis and treatment of celiac disease and the health risks associated with it. The fourth chapter is devoted to gluten-free foods, their labeling, basic rules for buying gluten-free foods and preparing gluten-free food. The fifth chapter is focused on the contributions of health insurance companies to gluten-free food. The following chapter deals with the hospitalization of a celiac in a medical facility and provides some tips for hospitalization. The last chapter of the theoretical part deals with education, the role of the general nurse in the education of patients with celiac disease, forms of education and lists the most common myths associated with celiac disease. In the empirical part, the first goal was to map the awareness and knowledge of general nurses about celiac disease, its symptoms, manifestations and treatment. The second goal was to map the specifics of nursing care for patients with celiac disease. Method of achieving the goal: The set goals were achieved through a research survey, which was carried out using a qualitative approach, the method of questioning. The method of semi-structured interview was chosen for data collection. The examined group consisted of eight general nurses working in inpatient departments at the Tábor Hospital, a.s. Scientific benefits of the work: The research shows that general nurses have awareness and knowledge of celiac disease, its symptoms, manifestations and treatment, although the answers to some questions were only partial, sometimes inaccurate and in minimal cases incorrect. The identified specifics of nursing care for celiacs can be used in hospital facilities to improve the quality of care provided. Furthermore, the work can be used by people who are interested in obtaining information related to celiac disease and a gluten-free diet. Acquired knowledge and conclusions: The research survey revealed the specifics of nursing care for people with celiac disease, which are mainly associated with the gluten-free diet provided. Furthermore, celiac disease has been found to be incorrectly associated with allergies.
Quality of patients life with celiac disease

This thesis is dealing with the quality of life of patients with celiac disease. In the theoretical part, I am presenting the characteristics of celiac disease and I am describing its history briefly. For better orientation in the issue, I also joined a description of the anatomy of the small intestine and pathophysiology of celiac disease. In this thesis I am also dealing with incidence of disease, etiology and risk factors participating in the development of celiac disease, symptoms and the different forms, which are related to celiac disease, as well as potential complications and prognosis of celiac disease. An irreplaceable is the role of nurses in diagnosis and targeted screening for celiac disease, therapy and prevention, dispensarization and education of patients with celiac disease about gluten-free diet. I am also describing the problems that patients with celiac disease may encounter in everyday life. Last but not least I am focusing on the general concept of quality of life and also on the quality of life related to health and celiac disease. The effect of gluten-free diet on β-cell residual capacity, immune function and gut microbiome in children with newly diagnosed type 1. diabetes Abstract The pathophysiology of the onset and progression of type 1 diabetes (T1D) is not fully understood. Gluten has a proinflammatory effect on the immune system and is therefore considered as one of the factors affecting the onset and progression of T1D. The aim of the thesis is to allow a complex insight into the role of the GFD on the residual β-cell capacity, T1D control, gut microbiome, gut permeability, subtypes of immune cells and the effect of gut microbiome transfer into germ-free non-obese diabetic (NOD) mice on the incidence of diabetes. On the group of 45 children with T1D (26 intervention group, 19 control group) we proved the association of the GFD with slower decrease of β-cell residual capacity (the difference in the trend of C-peptide decrease 409 pmol/l/year; p = 0,04) and lower HbA1c (by 7,8 mmol/mol; p=0,02). We also described the changes in the gut bacteria that were differentially abundant after the administration of the GFD and the changes in abundance of the regulatory and effector immune cells. Comparison of the options and financial demands of a regular diet with gluten-free diet

In the Czech Republic, 50 000 people are diagnosed with Celiac disease and the only known treatment for it is a gluten-free diet. For such patients, gluten consumption can provoke inflammation of a small bowel that can result in heavy consequences. Gluten is a protein contained in wheat, rye, barley, and oats and plays an important role in food production, because of its properties. Bakery and other gluten-free products are produced from other types of cereals. They also require a special technological process, separate spaces, and have a much smaller target group of consumers, which can increase their price at the end. People following a gluten- free diet have to be very careful while eating in restaurants and canteens because even a small amount of gluten can harm their health. Health insurance companies in the Czech Republic contribute various amounts to the GFD, however, mostly only for children. The goal of the research was to find out whether and for how much gluten-free food is more expensive than the ordinary one; and how the availability of gluten-free products and meals is assessed by people who have to eliminate gluten due to health reasons.
